Today I faced with very strange issue. I was creating some templates and contents on Umbraco. Suddenly, when I referesh my the page, I saw that all the contents became unpublished (with # url) and I could not browse any pages!
After re-publishing entire tree node, it became fine but since the web site is huge it took me 2 hours! Is there any information about this issue?
No, I only created a very simple content. I could not re-produce this issue! Since I re-publish it again now umbraco.config is there. That moment I did not check!

Are you running any custom code that accesses the Umbraco API?
It seems strange that all your content would suddenly unpublish itself. Check if the "/App_Data/umbraco.config" file is there, and has content in it.
