Is it possible to define Multi-Node Tree Pickers with XPath Expressions to nodes which the user has no access to?
Or is there a work around?

I believe that the MNTP's content (and media) tree does implement user-acccess/security settings. If an XPath was used to select restricted nodes, then MNTP would either not display them, or throw an error (I haven't tried this personally).
Not sure about having a workaround for this - could be worth raising an ticket on the issue tracker? http://issues.umbraco.org/issues/U4
