No problem with large original images, or generating large images with ImageGen.
Do bear in mind that you will need available memory on the server and massive dimensions take a lot of memory. Any chance your machine is running out of memory? You might see a message in the event viewer, for instance.
Are you using the patch for ImageGen? That has a couple fixes that would help if you're not already using it.
thumbs of large source images not showing
I have a problem when using large source jpgs, 2500+ pixels wide.
The thumb is showing a missing image icon on the page. Is there a max limit file size og dimension i should be aware of, or is there a solution.
im using imagegen for a press area, so large source images is kinda necessary...

Yeah ive installed the update.
Ive no resized the source images to 2500 pixels max width. And tha does the trick. ill look deeper into it when i get time.
Are you by any chance using IE?
I had a problem with images uploaded to umbraco not showing up in IE, but in FF and Chrome everything was fine.
(Tried with and without ImageGen)
My solution was to reupload the image with another nodename.
