I've added the hostnames and selected the different languages for each, but when I go to the relevant page it throws a 404 not found page and also on the root pages en.mysite.com, it resolves back to the hosting providers holding page.

Have you also added the bindings in IIS? What version of Umbraco are you currently using?

Thanks for the reply, the site is on a shared hosting platform, so not sure I'll have access to the bindings, I'll check.
