Could you please provide much more detail about your setup? :) As Charlie is pointing out above it seems like your database credentials are wrong. But I'm not sure wether the database has been created on your server or not?
Are you trying to get up and running on a local environment? Or are you trying to deploy to a live environment?
Make sure that you read the documentation on how to install Umbraco - If you dig through it you can find information about system requirements and how to setup permissions as well.

Is this becasue I dont have the original pwd and username, I took a db.bak and imported onto the sql express 2008 r2 and then added permission to the .\administrator (dbo).

Are you able to connect to the database in SQL management studio using those creditials? Does the user have the permissions to access the database?
The problem is the creditials you have are not valid for the SQL instance or the user does not have permissions on the database.

Ive added the .\administrator as a owner in the db permissions, do I need to go to into a table and change the the credentials ?

Ok what i would do i go into SQL management studio and try to connect from there first off.
If you have the ip address that will be the serverName. Do you know if you have a user that has SQL Server Authentication on the database?
In SQL Management studio:
Server=SERVERNAME\SQLEXPRESS;
Authentication: SQLSERVERAUTHENTICATION
USERNAME:db-01
PASSWORd: Password for the database
Once you can log into the database doing that try those settings in the web config.
If that does still does not work it will be because the user does not have sufficent permissions for the database which you will need to apply :)
