The esiest way is probably to uninstall and reinstall if you are ok with loosing the old data. There are some changes to the db so if you need to upgrade you'll have to follow the steps from support.newsletterstudio.org.
I've just installed the new version without uninstalling the old and bar a couple of issues it seems to have worked.
In Marcus' post in the thread Dennis linked to he mentions there may be an issue with settings getting lost and the SenderName field. And there were.
Both were easily solved though, I copied the settings from the live site and just manually added the SenderName field.
Another issue that I seem to have is that the license doesn't seem to be accepted anymore. It's showing as a trial version even though we have a license for the site.
Gonna make this another post as I think we may have posted at the same time.

Upgrading 1.3 to 1.4.1
I've been upgrading an old umbraco site to use the current version of umbraco and am now going through the installed packages to do the same.
What is the best way to upgrade newsletter studio though. Is it OK to just install the new version and it will overwrite the existing.
Hi Suzyb,
Maybe this post from Markus, can help you to find out how to upgrade from 1.3 to 1.4.1

Hi!
Whats the name of the lic-file you have in the bin-folder? It has to be named "NewsletterStudio.lic".
The license should work even after your upgrade.
